Brief description of study

PURPOSE: The overall objective of the project is to perform comparative metabolomics on frail and non-frail patients undergoing major surgery. We hypothesize that frail patients will have distinct baselines, suffer greater metabolic derangements, and recover more slowly than non-frail patients. Additionally, we hypothesize that those patients who undergo prehab will have fewer derangements and greater recovery than those frail patients who do not undergo prehab. The primary outcome variables are plasma metabolite levels obtained at baseline, after prehabilitation, and at two postoperative timepoints. Metabolomic assays will be performed by the metabolomics core and metabolites identified and annotated by the core. Groups will be compared across time points and non-frail and frail individuals will be compared. Metabolomic data will be subjected to standard secondary analysis including principal component analysis and enrichment analyses to identify specific pathways of interest. Skeletal muscle samples obtained at surgery will be preserved for targeted studies, corroboration of plasma findings, microscopy, and physiologic analysis.
